微生物来源天然产物中吡啶环结构单元的生物合成研究进展

摘 要: 吡啶环及其衍生结构作为骨架构造单元赋予了天然产物良好的生物活性。许多微生物来源的天然产物,特别是生物碱类及硫肽类化合物,大多含有吡啶环结构,其生物合成机制研究不仅可以丰富人们对天然产物组装逻辑的理解,还对开发新型抗生素具有显著意义。吡啶环生物合成机制可以概括为以下三类:1)赖氨酸 经由氨基转移酶环化脱氨形成吡啶环;2)[4+2]环化加成形成吡啶环;3)非酶催化形成吡啶环。本综述将介绍近年来一些代表性微生物来源天然产物中有关吡啶环生物合成研究的进展,并着重阐述蕴含其中的不同组装机制。

Title: Research progress on biosynthesis of pyridine ring unit contained in microbial natural products

Abstract: As one of the important skeleton building units for natural products assembly, the pyridine ring and its derivatives are usually conferred with excellent biological activities. Many microbial natural products, especially alkaloids and thiopeptides compounds, contain pyridine ring structures. Studies on the biosynthesis mechanisms of pyridine ring can not only enrich our understanding of natural product assembly logic, but also inspire the discovery of novel drugs significatively. The biosynthesis mechanisms of pyridine ring contained in microbial natural products can be summarized as follows: 1) the cyclodeamination of lysine catalyzed by specific aminotransferases; 2) [4+2] cyclic addition reactions; 3) non-enzymatic reactions. This review summarizes the recent research progress on the biosynthesis of pyridine ring in some representative microbial natural products, and emphasizes on the different assembly mechanisms of this interesting unit.
