基于rGO/Hemin/AuNPs复合材料的电化学传感器检测DNA

摘 要: 利用一锅合成法,在碱性环境且不添加任何表面活性剂、还原剂和稳定剂的条件下,通过加热氧化石墨烯(GO)、氯化血红素(Hemin)、氯金酸(HAuCl4)的混合溶液,合成了还原氧化石墨烯/Hemin/金纳米颗粒(rGO-H-Au)复合材料。该复合材料中GO被还原为rGO,rGO作为支持材料,首先克服了Hemin不易溶于水的缺点,其次抑制了AuNPs的聚集。正常条件下,该复合材料具有催化H2O2分解的作用,而单链DNA中的碱基可以与rGO发生π-π作用,抑制其催化H2O2分解的活性。目标物存在时,该DNA与目标物结合,从rGO-H-Au复合材料表面脱离,使rGO-H-Au复合材料的催化活性恢复。当检测特定DNA链时,将与rGO-H-Au复合材料结合的DNA设计成其互补链即可。基于此,构建了一种基于rGO-H-Au复合材料的电化学传感器,用于电化学检测DNA.

Title: A novel electrochemical sensor based on rGO/Hemin/AuNPs composites to detect DNA

Abstract: Using one-pot strategy, ternary reduced graphene oxide-Hemin-Au (rGO-H-Au) nanohybrids is synthsized by simply heating the mixed solution of GO, Hemin and HAuCl4 under alkaline conditions, without adding any additional surfactant, reducing agents and stabilizing agent. In this composite, GO was reduced to rGO. rGO acts as a supporting material, which overcomes the disadvantages of Hemin insoluble in water and inhibites the aggregation of AuNPs. Under normal conditions, the composite has the effect of catalyzing H2O2 decomposition, while the bases in single-stranded DNA (ssDNA) can interact with rGO through π-π stacking and inhibit the activity of the composite to catalyze H2O2 decomposition. While in the presence of targets, the single-stranded DNA combines with the targets away from the surface of the rGO-H-Au composite, and the catalytic activity of rGO-H-Au composite recovers. When specific DNA strands are detected, the DNA strand that binds to the rGO-H-Au composite could be replaced with its complementary strand. Based on this, a novel electrochemical sensor based on rGO-H-Au composite has been constructed for detecting DNA.
