金纳米颗粒在脂质微气泡上的组装研究

摘 要: 脂质微气泡被广泛应用于对比增强超声造影,胶体金纳米颗粒具有粒径可控、结构稳定和生物相容性良好的优点,近年来被应用于药物输运、光声成像和计算机断层扫描(computed tomography,CT)成像。本文将金纳米颗粒(23 nm)自组装到超声造影剂脂质微气泡膜壳上,实验采用真空旋转蒸发水化的方法制备了膜壳表面组装不同浓度金纳米颗粒的微气泡。通过扫描电子显微镜(scanning electron microscopy,SEM)和透射电子显微镜(transmission electron microscopy,TEM)等对金纳米颗粒和微气泡进行了表征。最后,验证了携载金纳米颗粒的微气泡的超声造影和CT造影增强的双模态造影增强的功能效果。

Title: Study of assembly on lipid-shelled microbubbles with gold nanoparticles

Abstract: Lipid-shelled microbubbles are widely used in contrast-enhanced ultrasound imaging. And colloidal gold nanoparticles have advantages of the controllable size, stable structure and good biocompatibility. In recent years, the gold nanoparticles have also been explored to be applied in drug delivery, photoacoustic imaging and computed tomography (CT) imaging. In this study, the 23 nm gold nanoparticles have been self-assembled on the shell of lipid-shelled microbubbles in ultrasound contrast agent. The microbubbles assembled with different concentrations of gold nanoparticles are prepared by vacuum rotary evaporation hydration method. The gold nanoparticles and microbubbles have been characterized by scanning electron microscopy (SEM) and transmission electron microscopy (TEM). At last, the function effect of dual contrast-enhanced ultrasound imaging and CT imaging in nanoparticle-assembled microbubbles has been further confirmed.
