氨基酸及其水合物的太赫兹光谱识别研究

摘 要: 太赫兹(terahertz,THz)光谱对分子的集体振动模式和分子间弱相互作用非常敏感,因此可以根据其吸收谱的差异实现分子无水及水合状态的识别。利用太赫兹时域光谱(THz time-domain spectroscopy,THz-TDS)技术分别对4种极性不同的氨基酸[甘氨酸(Gly)、L-丙氨酸(L-Ala)、L-丝氨酸(L-Ser)、L-酪氨酸(L-Tyr)]的无水和水合形式进行检测,发现各无水氨基酸及其水合物的太赫兹吸收谱图均存在明显差异,且差异程度与氨基酸亲水性指数呈正相关。结合傅里叶变换红外光谱(Fourier transform infrared spectroscopy,FTIR)、热重(thermogravimetry,TG)分析和X 射线衍射(X-ray diffraction,XRD)技术从分子基团、热力学及晶体结构角度对这4种氨基酸进行表征分析,以进一步验证太赫兹光谱的识别优越性及实验结果的准确性。

Title: Identification of amino acids and their hydrates by terahertz spectroscopy

Abstract: Terahertz (THz) spectroscopy is very sensitive to the collective vibrational mode of molecules and weak interaction between molecules. Therefore, the identification of anhydrous and hydration states of molecular can be realized according to the difference of terahertz absorption spectra. In this paper, the technique of THz time-domain spectroscopy (THz-TDS) was used to detect the anhydrous and hydrated forms of four kinds of amino acids (glycine, L-alanine, L-serine, L-tyrosine) owning different polarities. Based on the results mentioned above, we find that there is a significant difference between the absorption spectrums of amino acids and corresponding their hydrate, and the degree of difference mentioned above is positively correlated with the amino acid hydrophilicity index. Furthermore, in order to verify the recognition superiority and accuracy of experimental results of terahertz spectroscopy, Fourier transform infrared spectroscopy (FTIR), thermogravimetry (TG) analysis and X-ray diffraction (XRD) techniques were used to characterize and analyze the four kinds of amino acid from perspective of molecular groups, thermodynamic properties and crystal structure.
