三种植物油对高脂饮食诱导的小鼠肥胖和糖脂调节作用的比较

摘 要: 目的:探究冷榨椰子油对高脂饮食诱导小鼠的体重、能量代谢和糖脂代谢的影响。方法:将48只雄性C57BL/6J小鼠随机分成4组,即正常对照(ND)组、高脂椰子油(CO)组、高脂大豆油(SO)组和高脂米糠油(RO)组。每天上午喂食并记录采食量,每周监测小鼠体重和空腹血糖(fasting blood glucose,FBG)。高脂饲料干预的第12周进行葡萄糖耐量实验(glucose tolerance test,GTT),第15周用CLAMS动物全面代谢监测系统测定小鼠的呼吸交换率(respiratory exchange rate,RER)、自发活动量和能量消耗,第16周处死小鼠,用试剂盒测定血清生化指标[甘油三酯(triglyceride,TG)、总胆固醇(total cholesterol,TC)、高密度脂蛋白胆固醇( high density lipoprotein-cholesterol , HDL-c )、低密度脂蛋白胆固醇( low densitylipoprotein-cholesterol,LDL-c)、游离脂肪酸(free fatty acid,FFA)],用实时荧光定量PCR(quantitative real-time PCR,qRT-PCR)测定与脂代谢相关基因的表达水平。结果:与SO和RO组相比,CO组小鼠的体重、FBG、TG和FFA均显著降低(P<0.05),表现出更好的葡萄糖耐受性(P<0.05),自发活动量和能量消耗也明显增多(P<0.05),qRT-PCR结果表明肝脏SEEBP-1c、FAS和ACC-1的表达显著下调(P<0.05),PPARα、CPT1和CYP7A1的表达显著上调(P<0.05)。结论:与大豆油和米糠油相比,椰子油可以降低小鼠体重,增加自发活动量和能量消耗,抑制肝脏脂质的合成并加速氧化分解,改善胰岛素的敏感性。

Title: Comparison of three vegetable oils on obesity and glycolipid metabolism in mice induced by high-fat diet

Abstract: Objective: To investigate the effects of cold-pressing coconut oil on body weight, energy metabolism and glycolipid metabolism in mice induced by high-fat diet. Methods: Forty-eight male C57BL/6J mice were randomly divided into 4 groups, normal control (ND) group, high-fat coconut oil (CO) group, high-fat soybean oil (SO) group and high-fat rice bran oil (RO) group. Mice were fed every morning and their food intake was recorded. Body weight and fasting blood glucose (FBG) were monitored weekly, the glucose tolerance test (GTT) was performed at the 12th week of the high-fat diet intervention, and the respiratory exchange rate (RER), spontaneous activity and energy expenditure of the mice were measured at the 15th week using the CLAMS animal comprehensive metabolic monitoring system. The serum levels of triglyceride (TG), total cholesterol (TC), high density lipoprotein-cholesterol (HDL-c), low density lipoprotein-cholesterol (LDL-c) and free fatty acid (FFA) were measured after mice were sacrificed at the 16th week. The expressions of lipid metabolism-related genes in liver were analyzed by quantitative real-time PCR (qRT-PCR). Results: Compared with the SO and RO groups, the body weight, FBG, TG and FFA of the CO group were significantly lower (P<0.05), glucose tolerance was improved (P<0.05), spontaneous activity and energy expenditure also increased (P<0.05). qRT-PCR results showed that the expression of SEEBP-1c, FAS and ACC-1 in liver were significantly down-regulated (P<0.05), however, PPARα, CPT1 and CYP7A1 were significantly up-regulated (P<0.05). Conclusion: Compared with soybean oil and rice bran oil, coconut oil can reduce the mice’s body weight, increase spontaneous activity and energy expenditure, inhibit liver lipid synthesis and accelerate oxygenolysis, and improve insulin sensitivity.
