乳杆菌生物膜成膜规律及特性研究

摘 要: 采用经典的96孔板法测定了8个菌种35株不同乳杆菌的成膜能力,并研究不同成膜能力菌株的成膜规律和影响因素。结果显示,乳杆菌的成膜能力呈现菌株特异性,与菌种及菌株来源无相关性;扫描电子显微镜(scanning electron microscope,SEM)结果显示,乳杆菌可以在非水溶性膳食纤维表面形成特定结构的生物膜;葡萄糖、牛肉膏、培养基浓度对菌株的成膜能力存在显著影响,但无特定规律;菌体表面疏水性与菌株成膜能力呈正相关。此外,耐酸耐胆盐实验显示,膜样乳杆菌的抗逆性显著提高。综上所述,生物膜样化在乳杆菌产品的开发应用中具有非常大的潜在价值。

Title: Research on the regulation of Lactobacillus biofilm formation and the characteristics

Abstract: Classical method of 96-well plates was used to cultivate biofilms of 35 Lactobacillus strains from 8 species, finding out the regulations and influence factors of the strains with different biofilm-forming capability. Results showed that the ability of biofilm formation of these Lactobacillus strains was strain-specific, which means there was no relationship between the capacity and species or source. The pictures of scanning electron microscope (SEM) illustrated that Lactobacillus can form biofilms with specific structures on the surface of insoluble dietary fiber. Besides, the concentration of glucose, beef extract and medium had significant influence on the biofilm-forming capability, but they had no specific rules. However, the surface hydrophobicity of strains was positively correlated with their biofilm-forming capability. In addition, acid and cholate resistance test result indicated that the resistance of biofilm-like Lactobacillus was significantly improved. To sum up, it showed a large potential of the application of Lactobacillus biofilms in the probiotics production.
