小鼠急性心肌梗死模型的建立及心功能评价

摘 要: 目的:通过结扎冠状动脉左前降支建立小鼠急性心肌梗死(acute myocardial infarction,AMI)模型,并使用心脏超声进行心功能评价分析。方法:C57BL/6雄性小鼠30只,进行小动物无创气体麻醉,采用左侧肋间手术路径进入左胸腔,结扎冠状动脉左前降支,建立AMI模型。术后3周使用小动物超声影像系统评价小鼠的左室收缩末期直径(left ventricular end systolic diameter,LVESD)、左室舒张末期直径(left ventricular end diastolic diameter,LVEDD)、射血分数(ejection fraction,EF)和缩短分数(fractional shortening,FS)。超声检查完成后取心脏标本进行病理学检查。结果:小鼠建模术后3周内死亡率为13.3%(4/30),使用超声检查评价,建模成功率为86.7%(26/30). 建模后小鼠心功能(EF/FS)明显下降,心室(LVESD 和LVEDD)明显增大,病理切片可见大量心肌细胞坏死,瘢痕形成。结论:采用结扎冠状动脉左前降支可以成功建立小鼠AMI模型,并可用无创超声评价心功能。

Title: Establishment of murine model for acute myocardial infarction and assessment of cardiac function

Abstract: Objective: To establish a murine model of acute myocardial infarction (AMI) by ligating the left anterior descending coronary artery and to assess cardiac function by echocardiography. Methods: Thirty C57BL/6 male mice were used in this study. Into the left thoracic cavity by the left intercostal surgery route, the left anterior descending coronary artery was ligated after noninvasive anesthesia, and then the AMI model was established. The echocardiographic assessment of myocardial infarction and cardiac function was performed after operation 3 weeks, the pathological examination of the heart specimens was performed after echocardiography. Left ventricular end systolic diameter (LVESD), left ventricular end diastolic diameter (LVEDD), ejection fraction (EF) and fractional shortening (FS) were measured and analyzed. Results: The acute mortality was 13.3% (4/30) after establishing the murine model for 3 weeks. The echocardiography showed that the success rate of establishment of the infarction model was 86.7% (26/30). Cardiac function marker EF/FS decreased significantly, ventricular size marker LVESD and LVEDD increased significantly. Pathological examination confirmed the necrosis of cardiac myocytes and the scar formation. Conclusion: The murine AMI model was successfully established by ligation of left anterior descending coronary artery, and noninvasive echocardiography could be used to assess cardiac function.
