鞣花酸和尿石素A对高脂膳食诱导肥胖小鼠血脂及肠道菌群的影响

摘 要: 目的:比较鞣花酸(ellagic acid,EA)和尿石素A(urolithin A,UroA)影响高脂膳食诱导肥胖小鼠血脂及肠道菌群的差异。方法:雄性C57BL/6J小鼠用普通饲料和高脂饲料饲喂9周,选取高脂饲料组肥胖小鼠,分别用低剂量(10 mg⋅kg−1⋅d−1)、高剂量(30 mg⋅kg−1⋅d−1)EA和UroA(20 mg⋅kg−1⋅d−1)灌胃处理7周,测体重、体脂、肝重、代谢活动、血脂及血清抗氧化指标,并用16S rRNA基因扩增子高通量测序分析结肠内容物菌群构成。结果:EA高、低剂量组及UroA组均能不同程度地降低高脂饮食诱导小鼠的脂肪指数,增加其夜间活动量,提高血清高密度脂蛋白胆固醇(high density lipoprotein cholesterol,HDL-C),降低血清丙二醛(malondialdehyde,MDA),提高血清超氧化物歧化酶(superoxide dismutase,SOD),但低剂量EA作用更明显。在属水平上,肥胖小鼠结肠菌群Desulfovibrio 显著升高(P<0.01),而各处理组均可降低其含量,UroA组具有显著性差异(P<0.05);各处理组可不同程度地增加Odoribacter、Coprostanoligenes、Xylanophilum、Ruminococcaceae_UCG-010和Subdoligranulum含量。结论:高、低剂量EA和UroA均可改善高脂饮食诱导肥胖小鼠的血脂水平并调节其肠道菌群,但低剂量EA的效果最好。

Title: Effects of ellagic acid, urolithin A on serum lipids and intestinal flora in the fat mice induced by high-fat diet

Abstract: Objective: To compare the differences in the effects of ellagic acid (EA), urolithin A (UroA) on serum lipids and intestinal flora in fat mice induced by high-fat diet. Methods: Male C57BL/6J mice were fed normal diet and high-fat diet for 9 weeks, then fat mice in the high-fat diet group were selected and gavaged with low dose (10 mg⋅kg−1⋅d−1), high dose (30 mg⋅kg−1⋅d−1) of EA and UroA (20 mg⋅kg−1⋅d−1) respectively for 7 weeks. Body weight, body fat, liver weight, metabolic activity, serum lipids and antioxidant indexes were measured, and 16S rRNA gene amplifiers were used for high-throughput sequencing to analyze the microbiota of colonic contents. Results: Both the high and low dose groups of EA and the UroA group decreased body fat rate, increased their nocturnal activity, increased serum high-density lipoprotein cholesterol (HDL-C), decreased serum malondialdehyde (MDA), and increased serum superoxide dismutase (SOD), but the effect of low dose of EA was more obvious. At genus level, the colon microbiota Desulfovibrio significantly increased in fat mice (P<0.01), while each treatment group could reduce its content, and UroA group showed significant difference (P<0.05). Each treatment group could increase the content of Odoribacter, Coprostanoligenes, Xylanophilum, Ruminococcaceae_UCG-010 and Subdoligranulum in different degrees. Conclusion: Both high and low doses of EA and UroA could improve the serum lipids and regulate the intestinal flora of fat mice induced by high-fat diet, but the effect was best in the low dose of EA.
