预先低剂量微波辐射增强γ射线诱导小鼠白血病L1210细胞凋亡

摘 要: 目的:研究预先低剂量微波辐射对γ射线诱导L1210细胞凋亡的影响。方法:将小鼠白血病L1210细胞随机分为对照组(C)、单纯微波组(RF)、单纯γ射线组(γ)和联合暴露组(RF+γ)。单纯微波组和联合暴露组给予120 μW/cm2微波辐射,4 h/d,连续5 d;第5天给予单纯γ射线组和联合暴露组8 Gy γ射线照射。用钙离子依赖性磷脂结合蛋白-异硫氰酸荧光素/碘化丙啶(Annexin V-FITC/PI)双标记方法检测细胞凋亡,通过Western blotting检测Survivin蛋白的表达水平。结果:与对照组相比,单纯γ射线组凋亡率显著升高(P<0.05);与单纯γ射线组相比,联合暴露组凋亡率显著升高(P<0.05)。与对照组相比,单纯γ射线组细胞内Survivin蛋白表达水平显著降低(P<0.05);与单纯γ射线组相比,联合暴露组Survivin蛋白表达水平显著降低(P<0.05)。结论:在本实验条件下,8 Gy γ射线可以引起小鼠白血病L1210细胞凋亡率升高,Survivin蛋白表达水平降低。预先900 MHz、120 μW/cm2微波辐射能够显著增强γ射线引起的小鼠白血病L1210细胞的损伤,表现为细胞凋亡率升高和Survivin蛋白表达水平降低。

Title: Apoptosis of mouse leukemia L1210 cells induced by pre-low-dose microwave radiation-enhanced γ-rays

Abstract: Objective: To investigate the role of pre-low-dose microwave radiation on γ-rays inducing the apoptosis of L1210 cell. Methods: L1210 cells were radomly divided into control group (C), microwave alone group (RF), γ-rays alone group (γ) and combined exposure group (RF+γ). Microwave alone group and combined exposure group were exposed to 120 μW/cm2 microwave, 4 h/d, for 5 days. The combined exposure group and γ-rays alone group were subsequently exposed to 8 Gy γ-rays on 5th day. Cell apoptosis was detected with Annexin V-FITC/PI double labelling method. Survivin protein expression level was measured with Western blotting. Results: Compared with control group, the apoptosis rate of cells in γ-rays alone group significantly increased (P<0.05). The apoptisis rate of cells in combined exposure group was statistically higher than that of γ-rays alone group (P<0.05). Compared with control group, Surviving protein expression level of γ-rays alone group decreased significally (P<0.05). Survivn protein expression level of combined exposure group was statistically lower than that of γ-rays alone group (P<0.05). Conclusion: Under this experimental condition, 8 Gy γ-rays could increase the apoptosis rate and decreased Survivin protein expression level in L1210 cells. Pre-exposure with 900 MHz, 120 μW/cm2 microwave radiation could significantly increase the γ-rays induced damages to L1210 cells, as evidenced by the increase of apoptosis rate and decrease of Survivin protein expression level.
