酶法合成油酸龙脑酯

摘 要: 为提高龙脑的稳定性和生物利用度,本文研究了油酸龙脑酯的合成工艺。以右旋龙脑和油酸为原料,在无溶剂体系中,酶法合成油酸龙脑酯。以油酸龙脑酯的酯化率为指标,探究了脂肪酶种类、反应温度、底物物质的量比、酶添加量和反应时间对酯化率的影响。确定在无溶剂体系下合成油酸龙脑酯的最优工艺条件:催化剂为AYS脂肪酶,AYS添加量为8%(以底物总质量计),反应温度为45℃,底物(右旋龙脑与油酸)物质的量比为1:2,反应时间为48 h,在此条件下产物的酯化率为96.54%. 通过分子蒸馏和酸碱中和纯化产物,并通过质谱和核磁对产物进行结构鉴定。结果显示:确认产物为油酸龙脑酯,产物纯度达到97.51%.

Title: Enzymatic synthesis of bornyloleate

Abstract: In order to improve the stability and bioavailability of borneol, the synthetic process of bornyloleate was studied in this paper. d-borneol and oleic acid were used as raw materials to synthesize bornyloleate enzymatically in solvent-free system. The effects of lipase type, reaction temperature, substrate molar ratio, lipase load and reaction time on the esterification degree were investigated, which the esterification degree of bornyloleate was taken as the index. The optimal conditions of synthesis of bornyloleate in solvent-free system were obtained as follows: AYS lipase was as catalyst and AYS lipase load was 8% (based on total mass of substrates), the reaction temperature was 45℃, the molar ratio of d-borneol to oleic acid was 1:2 and the reaction time was 48 h. Under these conditions, the esterification degree of the reaction product was obtained with 96.54%. The product was purified by molecular distillation and acid-alkali neutralization, and the structure of the product was identified subsequently by mass spectrometry and nuclear magnetic resonance. The results showed that the product was confirmed to be bornyloleate and the purity of that was 97.51%.
