硫酸化壳聚糖增强型水凝胶促进BMP-2诱导MC3T3-E1生长研究

摘 要: 本文研究了三种不同硫酸化壳聚糖(sulfated chitosan,SCS)在水凝胶中调节骨形态发生蛋白-2(bone morphogenetic protein-2,BMP-2)活性,并诱导小鼠胚胎成骨细胞前体细胞(MC3T3-E1)生长及成骨的作用。首先制备了三种具有不同硫酸化结构的SCS,并通过傅里叶变换红外光谱(Fourier transform infrared spectroscopy,FTIR)、核磁共振氢谱(1H nuclear magnetic resonance spectroscopy,1H NMR)和元素分析进行了结构确认。然后将SCS与壳聚糖(CS)-海藻酸钠(sodium alginate,SA)复合制备成SCS增强型水凝胶,并对材料的流变学性能及微观形貌进行了分析。最后在体外评价了载有BMP-2的三种水凝胶中MC3T3-E1细胞的生长情况及成骨能力。结果表明,SCS增强型水凝胶能提高BMP-2活性,低剂量BMP-2(75 ng/mL)即可促进MC3T3-E1细胞生长及碱性磷酸酶(alkaline phosphatase,ALP)分泌。高度硫酸化的SCS具有最好的BMP-2促进作用,显示出较好的骨修复应用潜力。

Title: Sulfated chitosan enhanced hydrogel for promoting BMP-2 induced MC3T3-E1 growth

Abstract: In this study, the effects of three different sulfated chitosans (SCSs) on regulating the activity of bone morphogenetic protein-2 (BMP-2) and inducing cell growth and osteogenesis of mouse preosteoblast (MC3T3-E1) in hydrogels were investigated. Firstly, three SCSs with different sulfated structures were synthesized. Their chemical structures were confirmed by using Fourier transform infrared spectroscopy (FTIR), 1H nuclear magnetic resonance spectroscopy (1H NMR), and elemental analysis. Then, these SCSs and chitosan (CS)-sodium alginate (SA) were composited to prepare SCS enhanced hydrogels. The rheological properties and morphology of these hydrogels were analyzed. Finally, the growth and the osteogenic capacity of MC3T3-E1 cells in the hydrogels which loaded with BMP-2 were evaluated in vitro. The results showed that the SCS enhanced hydrogels could improve the activity of BMP-2. So, the growth of MC3T3-E1 cells and the secretion of alkaline phosphatase (ALP) cloud be promoted with a low dose of BMP-2 (75 ng/mL). It also showed that the highly sulfated SCS had the best function on BMP-2 enhancement, which had good application potential for bone repairing.
