桦褐孔菌对UVB诱导HaCaT细胞光老化的保护作用研究

摘 要: 目的:研究桦褐孔菌提取物对中波紫外线(UVB)诱导的人表皮角质形成细胞(HaCaT)光老化的修复及机制。方法:提取桦褐孔菌多糖组分、正己烷组分、乙酸乙酯组分、正丁醇组分,通过2,2-联苯基-1-苦基肼基(2,2-diphenyl-1-picrylhydrazyl,DPPH)法对不同提取物抗氧化活性进行检测。通过UVB辐射HaCaT细胞构建细胞光老化模型,评价不同提取物对细胞衰老及活性氧(reactive oxygen species,ROS)水平的影响,探讨其抗皮肤光老化的作用机制。结果:体外抗氧化活性检测结果显示,桦褐孔菌提取物具有良好的抗氧化活性,多糖组分的DPPH自由基半最大效应浓度(concentration for 50% of maximal effect,EC50)最低,具有最佳的抗氧化活性。细胞水平上结果显示,桦褐孔菌提取物明显降低UVB引起的细胞衰老及抑制ROS水平,其中多糖组分给药效果最佳。与模型组相比,桦褐孔菌提取物明显降低细胞内p16、p21和p53的表达,抑制MMP-1、MMP-3,促进Collagen І和Collagen Ш产生,证明桦褐孔菌提取物能够缓解UVB引起的细胞衰老。结论:桦褐孔菌提取物对UVB引起的HaCaT细胞光老化具有良好的缓解作用,多糖组分具有最佳的抗皮肤光老化活性,这种作用可能与桦褐孔菌提取物具有较好的抗氧化活性、抑制细胞衰老以及促进胶原蛋白产生相关。

Title: Intervention effect of Inonotus obliquus on UVB-induced photoaging in HaCaT cells

Abstract: Objective: To investigate the effect and mechanism of Inonotus obliquus extracts on UVB-induced photoaging in human epidermal keratinocytes (HaCaT). Methods: Different components of I. obliquus were extracted, including polysaccharide component, n-hexane component, ethyl acetate component and n-butanol component, and the antioxidant activity of different extracts was detected by 2,2-diphenyl-1-picrylhydrazyl (DPPH) method. The photoaging model of HaCaT cell was established by UVB radiation to evaluate the effects of different extracts on cell senescence and reactive oxygen species (ROS) activity, and the possible mechanism of anti-skin photoaging was finally explored. Results: The in vitro antioxidant activity test showed that the I. obliquus extracts had good antioxidant activity, the DPPH free radical concentration for 50% of maximal effect (EC50) of the polysaccharide component was the lowest, and it had the best antioxidant activity. At the cellular level, the results showed that the I. obliquus extracts significantly reduced UVB-induced cell senescence and inhibited ROS levels, and the polysaccharide component had the best effect. Compared with the model group, the I. obliquus extracts significantly reduced the expression of p16, p21 and p53 in the cells, inhibited MMP-1 and MMP-3, and promoted the production of Collagen І and Collagen Ш, which proved that the I. obliquus extracts alleviated the cell senescence caused by UVB. Conclusion: The I. obliquus extracts have a good effect on HaCaT cell photoaging caused by UVB, and the polysaccharide component has the best anti-skin photoaging activity. This effect may be related to the good antioxidant activity, the inhibition of cell senescence and the promotion of collagen production of the I. obliquus extracts.
