双发射三磷酸腺苷荧光探针的合成及应用

摘 要: 三磷酸腺苷(adenosine triphosphate,ATP)能在活细胞中产生和存储能量,并参与糖酵解、蛋白质的转运活性、克雷布(Kreb)循环等生命活动。溶酶体ATP浓度异常会导致溶酶体功能紊乱,因此溶酶体ATP水平可视化对探索与溶酶体ATP相关的生物过程具有重要意义。本文通过酰氯化、酰胺化反应构建了ATP荧光探针RP-ATP,探针以罗丹明B和芘甲酸为荧光团,二者由二亚乙基三胺通过酰胺键连接。ATP与探针结合后,促使芘甲酸和罗丹明B解聚集以及罗丹明B螺内酰胺环开环,显著增强荧光。探针对ATP的响应范围为毫摩尔级别,与ATP的生理浓度相匹配,且能在酸性pH范围内识别溶酶体内的ATP. 探针RP-ATP是探究与溶酶体ATP相关生理、病理过程中强有力的工具。

Title: Synthesis and application of dual-emission fluorescent probe for adenosine triphosphate

Abstract: Adenosine triphosphate (ATP) can produce and store energy in living cells, and participates in glycolysis, protein transport activity, Kreb cycle and other biological functions. Abnormal lysosomal ATP concentration can cause lysosomal dysfunction, and visualization of lysosomal ATP levels is of great significance for exploring the biological processes related to lysosomal ATP. In this paper, an ATP fluorescent probe RP-ATP was constructed through acylation and amidation reactions. The probe uses rhodamine B and pyrenecarboxylic acid as the fluorophores, which are connected by diethylene triamine through an amide bond. After ATP binds to the probe, it promotes the de-aggregation of pyrenecarboxylic acid and rhodamine B and the ring opening of the rhodamine B spirolactam ring. The fluorescence is significantly enhanced. The response range of the probe to ATP is in the millimolar level which matches the physiological concentration of ATP, and can recognize ATP in the lysosome in the acidic pH range. The probe RP-ATP can be a useful tool for exploring the physiological and pathological processes related to lysosomal ATP.
