IFN-γ联合TNF-α刺激间充质干细胞缓解顺铂诱导的肾纤维化

摘 要: | 目的:探讨间充质干细胞(mesenchymal stem cells,MSCs)以及干扰素-γ(interferon-γ,IFN-γ)联合肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)刺激的MSCs [(I+T)MSCs]在顺铂诱导肾纤维化中的作用。方法:通过流式细胞术鉴定人脐带来源MSCs的表面标记;建立顺铂诱导的小鼠肾纤维化模型并检测血清尿素氮(blood urea nitrogen,BUN)指标;实时定量PCR(real-time quantitative PCR,RTQ-PCR)检测(I+T)MSCs趋化因子和免疫抑制相关基因的表达以及肾损伤和纤维化相关基因的表达;利用病理染色评估肾损伤和肾纤维化水平;使用流式分析肾中CD45+细胞的浸润程度。结果:MSCs未能改善顺铂诱导的肾纤维化水平,但(I+T)MSCs可以缓解顺铂诱导的肾纤维化。结论:(I+T)MSCs发挥免疫调控功能,抑制炎症细胞浸润从而缓解顺铂诱导的肾纤维化。 |

Abstract: | Objective: To investigate the role of mesenchymal stem cells (MSCs) and interferon-γ (IFN-γ) combined with tumor necrosis factor-α (TNF-α) stimulated MSCs [(I+T) MSCs] in cisplatin-induced renal fibrosis. Methods: The surface markers of human umbilical cord-derived MSCs were identified by flow cytometry. A mouse model of renal fibrosis induced by cisplatin was established and the indexes of blood urea nitrogen (BUN) were detected. Real-time quantitative PCR (RTQ-PCR) was used to detect the expression of chemokines and immunosuppression-related genes in (I+T) MSCs and the expression of genes related to renal injury and fibrosis. Pathological staining was used to evaluate the level of renal injury and renal fibrosis. The infiltration of CD45+ cells in the kidney were measured by flow cytometry. Results: MSCs failed to ameliorate cisplatin-induced renal fibrosis, but (I+T) MSCs could alleviate cisplatin-induced renal fibrosis. Conclusion: (I+T) MSCs exert immunomodulatory function, which inhibits the infiltration of inflammatory cells to alleviate renal fibrosis induced by cisplatin. |
