三个小麦-长穗偃麦草矮秆种质系的鉴定和分析

摘 要: 长穗偃麦草(Thinopyrum elongatum,2n=70)是小麦重要的近缘植物之一。利用细胞学和原位杂交技术对三个小麦-长穗偃麦草后代选育的矮秆种质系进行鉴定,结果表明矮杆种质系山农31504染色体数目为44,细胞学稳定,属于二体异附加系;山农31504-1和山农31504-3染色体数目为42,细胞学稳定;以拟鹅冠草(Pseudoregnaria strigosa,St)总基因组DNA为探针进行基因组原位杂交(genomic in situ hybridization,GISH)鉴定,结果表明山农31504附加了2条长穗偃麦草St基因组染色体,山农31504-1和山农31504-3为小麦−长穗偃麦草小片段易位系;利用pTa535和pSc119.2为探针对山农31504-1、31504-3和鲁麦5号进行双色荧光原位杂交(fluorescence in situ hybridization,FISH),发现山农31504-1和31504-3的6B、1D染色体发生相同的结构变异,说明二者是相同的小麦-长穗偃麦草渐渗系,染色体结构变异可能发生在6B、1D染色体上。

Title: Identification and analysis of three Thinopyrum elongatum dwarf germplasm lines

Abstract: Thinopyrum elongatum (2n=70) is an important relative of wheat. We intended to identify three dwarf germplasm lines which were selected from filial generation of wheat-Th. elongatum by cytology and in situ hybridization. As the results showed, the chromosome number of Shannong 31504 was 44 and its cytology was stable, indicating a disomic alien addition line. Shannong 31504-1 and 31504-3 had 42 chromosomes and stable cytology. Genomic in situ hybridization (GISH) was preceded using genomic DNA from Pseudoregnaria strigosa (St) as the probe, concluding two alien chromosomes of Shannong 31504 belonged to St genome. Shannong 31504-1 and 31504-3 were wheat-Th. elongatum alien translocation with little fragment of Th. elongatum. Fluorescence in situ hybridization (FISH) with the probe of pTa535 and pSc119.2 was used to establish the genome constitution of Shannong 31504-1 and 31504-3, Lumai 5. We found that their 6B and 1D chromosomes had the same structural mutation. It indicated they were the same wheat-Th. elongatum introgression lines, and the chromosome structural mutation may occur on 6B and 1D chromosomes.
